Preclinical assessment of a novel small molecule inhibitor of tryptophan 2,3-dioxygenase 2 (TDO2)

چکیده
Introduction iTeos leverages the science of LICR to target metabolism of the tumor microenvironment and develops small molecule immunomodulators. TDO2 is a heme-containing enzyme highly expressed in the liver, in specific brain regions and liver/brain tumors. It catalyses the first and rate-limiting step of tryptophan catabolism along the kynurenine pathway and thereby regulates systemic tryptophan levels. Tryptophan (Trp) degradation by TDO2 suppresses anti-tumor immune responses and promotes tumor cell survival.

Preclinical assessment of a novel small molecule inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1)
Introduction iTeos leverages the science of the LICR to target the metabolism of the tumor microenvironment and develops small-molecule inhibitors. Tryptophan catabolism can suppress the anti-tumor immune response through expression of the rate-limiting enzyme IDO1. Local tryptophan reduction and metabolite production by the kynurenine pathway are associated with anergy/apoptosis of tumor-infil...
متن کاملIndoleamine 2,3-Dioxygenase and Immunological Tolerance during Pregnancy
Indoleamine 2,3-dioxygenase (IDO), an enzyme involved in the catabolism of tryptophan, is expressed by a variety of cells and tissues such as macrophages, dendritic cells, cells of the endocrine system and by the placenta. IFN- γ is the main inducer of this enzyme. IDO acts as an important defense mechanism of innate immunity against pathogens. It also has tumor suppressive activity and prolong...
متن کاملAssociation of tryptophan 2,3 dioxygenase gene polymorphism with autism.
Although elevation of blood and platelet serotonin has been documented in autism, genetic analyses of serotonin transporter gene have given conflicting results. Tryptophan 2,3 dioxygenase (TDO2) is the rate-limiting enzyme in the catabolism of tryptophan, the precursor of serotonin. A mutation that results in decreased activity of the TDO2 can decrease catabolism of tryptophan and increase the ...
متن کاملAccumulation of an Endogenous Tryptophan-Derived Metabolite in Colorectal and Breast Cancers
Tumor immune escape mechanisms are being regarded as suitable targets for tumor therapy. Among these, tryptophan catabolism plays a central role in creating an immunosuppressive environment, leading to tolerance to potentially immunogenic tumor antigens. Tryptophan catabolism is initiated by either indoleamine 2,3-dioxygenase (IDO-1/-2) or tryptophan 2,3-dioxygenase 2 (TDO2), resulting in biost...
متن کاملInduction of TDO2 and IDO2 in Liver by High-Fat Feeding in Mice: Discrepancies with Human Obesity
Low-grade and chronic inflammation is elicited in white adipose tissue in human obesity. The presence of inflammatory molecules leads to an increased tryptophan catabolism through the induction of indoleamine-2,3-dioxygenase-1 (IDO1). In order to characterize the mechanisms underlying this dysregulation, we have studied 2 mouse models of obesity. Unexpectedly, we did not detect any IDO1 express...
